Overview

Manifold Bio is a dynamic biotech company building a pipeline of targeted biologics using a novel in vivo-centric discovery approach. Utilizing massively parallel, in vivo screening and proprietary protein barcoding, the company drives the discovery of antibody-like drugs and other biologics.

Position

We are seeking an exceptional Associate Scientist with hands-on expertise in adeno-associated virus (AAV) vector production, purification, and analytical characterization to join our team. You will play a key role in generating high-quality, well-characterized AAV lots for rodent and non-human-primate (NHP) in vivo studies.

Responsibilities

  • Produce, purify, and qualify AAV vector lots from DNA to study-ready material.
  • Maintain and troubleshoot cell-culture systems and bioreactors.
  • Develop and execute QC assays; analyze and report data.
  • Drive process-optimization experiments to improve yield and quality.
  • Draft and maintain SOPs, batch records, and quality documents.
  • Collaborate with molecular biology, protein engineering, and external partners.

Requirements

Education & Experience: B.S. or M.S. in Biochemistry, Molecular Biology, Chemical Engineering, Virology, or related field with 2–5 years of industry experience. GLP/GMP experience preferred.

Wet-Lab Expertise: Hands-on experience with AAV production (HEK293 triple transfection and/or baculovirus-Sf9), downstream purification methods including affinity and ion-exchange chromatography, and tangential-flow filtration.

Analytical Expertise: Experience with qPCR/ddPCR, ELISA, HPLC, and other analytical assays. Ability to troubleshoot and interpret assay data.

Core Competencies: Attention to detail, excellent organization and communication skills, and a collaborative mindset.

Preferred Qualifications

Experience with large-scale AAV campaigns for NHP studies, familiarity with DoE, process characterization, GMP quality systems, and exposure to other viral vectors or lipid-nanoparticle formulation is a plus.
